I am using Filemaker 13 Advanced.

In the security setting I have "View only" for a particular layout. However, in this same layout I would like to make one field active, how do I do this?

First of all, security settings are not limited to "a particular layout".

Now, to make all fields in a table "view only" except for one field that should be modifiable by the users, chose Records: Custom privileges… then select the table and choose Field Access: limited…. You'll see a list of all fields in the selected table with the option to choose the permitted access for each.
