When I want to print an envelope I must change the printer setup options. Then it stays that default for all my layouts

How do I save the layout (such as an envelope) with those printer options

If you script the printing you can use the print setup(restore) scipt step to recall the details of any given job. The script can then be attached to a button on your layout
