Yes. You can limit the menu options available by using minimal custom menus, and you can override standard right-click menu items with your own. For example, create a menu set using a copy of the File menu. Edit the File menu so that the "Send mail..." option has a custom title, and set it to run a script. Associate this File menu with your custom menu for a layout, go to that layout, and right-click. You should see the custom menu item now, instead of "Send mail..."
