Closing Host

Can a script be initiated to close a host file while logged in from another box as a remote user. In order to make updates to a host pc i have to log in from a secondary machine but until the file is closed i cannot replace the file.

Hope i explained that well. My scripting so far closes the files on remote but not the host. Can't figure out how to initiate it on the host though?
